研究概要	大腸菌ゲノムの編制像を明らかにするため、転写開始と転写・翻訳制御カスケ-ド反応に関わる遺伝子群、tRNA遺伝子群、分泌系内膜構成遺伝子群、解糖系酵素遺伝子群、プリン生合成遺伝子群等の構造と機能の解析、並びに、それらの遺伝子がいかに編成、再編成されたかを他の生物種との比較を加えて解析した。まず、tRNA遺伝子の全体像を理解するため、これまで明らかにした全遺伝子の周辺部位の構造を解析した結果、幾つかのtRNA遺伝子間や遺伝子の下流には約200塩基対の反復配列が存在することを見いだし、これらが遺伝子の重複やゲノム上での分散に関わっていることが示唆された。転写開始因子σ54をコ-ドする<ntrA>___ー遺伝子領域を<P.putida>___ーの<ntrA>___ー遺伝子をプロ-ブとしてクロ-ン化し、その構造を解析すると共に、σ54を利用する遺伝子群を検索するためのベクタ-の作製をTolプラスミドシステムを用いて構築した。一方、RNAポリメラ-ゼβ'サブユニットと相互作用する転写終結因子NusAタンパク質の作用部位を<nusA>___-変異株を用いて調べたところ、アルギニン残基に富むドメインが重要であることが明かとなった。タンパク質の分泌に必須な<secY>___ー遺伝子の変異を抑制する<ssyB>___ーは<nusB>___ー遺伝子と同じであること、さらに、マルチコピ-サプレッサ-<msyA>___ーが核タンパク質HーNSをコ-ドする遺伝子であることを見いだした。大腸菌プリン生合成遺伝子はゲノム上に分散してレギュロンを構成しているが、枯草菌、コリネバクテリア菌との比較から、これらの遺伝子は最初に一つのオペロンとして編成されたものが、大腸菌では新しい制御システムを獲得しながら反復配列REP等を介して再編成されたことが示された。これまで明らかにした挿入因子<IS1>___ー、<IS2>___ー、<IS3>___ーに加えて、<IS5>___ー、<IS30>___ーのゲノム上の分布を検索し、W3110株においてはそれらが各々23、5コピ-存在することを見いだすと共に、<IS1>___ーには少なくとも4種のサブタイプが在ることを明らかにした。
