研究概要

コドン選択を決めている要因を網羅的に解析し、それらの因子間の相互関係を知り、コドン選択パタ-ンの持つ生物学的意味の総合的理解を目指している。池村はコドン3文字目が顕著にGCに偏るヒト遺伝子類がRバンドの内でも特に熱安定性の高いTバンドやテロメアから始まる最初のRバンドに集中する傾向を見い出し、体細胞キアズマを起し易い部位との関連も明らかにした。三田は温血動物のポリユビキチンとモノユビキチン遺伝子のコドン3文字目のGC%の顕著な差を見い出し、染色体バンド構造との関係を解析している。熊谷はコドン選択性とタンパク質合成効率の関係を放線菌遺伝子を対象に、実験的に証明した。陶山はコドン3文字目によるAT/GC使用の相殺調節がmRNA構造の安定性を通してタンパク質生産量に関係する可能性について、実験的検証を行っており、和田は相殺調節を含むコドン選択に関する一般法則の理論的研究を行った。小笠原は枯草菌の複製開始点領域の塩基配列を基礎に、コドン選択と遺伝子発現量との関係を明らかにした。武藤はゲノムGC%の顕著に異なる真正細菌類に着目し、それらの遺伝子構造を決定し、同義コドン置換がAT/GC圧の方向性を持って起っている事を明らかにした。五條堀はウイルス複製酵素のエラ-生成の性質の差がウイルスコドン選択パタ-ンに反映する事を示した。真木はGC/AT変異圧の実体に迫る研究として、A:TからC:Gへのトランスバ-ジョンに特異的なミュ-テ-タである大腸菌mutT遺伝子産物の生化学的解析を行いこのトランスバ-ジョンが、ヌクレオチドプ-ル中に含まれる微量の変異原性を持つ未知の誘導体に起因し、MutTタンパク質はこれを特異的に分解してエラ-生起を抑制しているとの結果を得た。
