研究概要	<ras>___ー蛋白を初めとする低分子量GTP結合蛋白は神経系に高含量に存在し、細胞の増殖・分化・小胞輸送などに必須の役割を果たしていると考えられている。我々は、ボツリヌス毒素由来のADPリボシル化酵素C_3がこの蛋白群の一つであるrho蛋白(Gb)を特異的に修飾すること、この修飾に伴いPCー12細胞などで神経突起形成などの神経分化様の変化が煮起されることを見出してきた。このことは、Gbが神経細胞の形態形成に一定の役割を果たしていることを示すものと考えられる。本研究ではこのような機能の発現に至る道筋を明らかにするため、この蛋白のGTP水解活性を調節する蛋白(GTPase活性化蛋白,GAP)を検索・同定し、均一まで純化した。〔方法〕Gbを既報の方法でウシ副賢可溶性分画より精製した。この蛋白のGTP水解活性を指標として、この活性を亢進する蛋白を副賢ホモジネ-トで同定し、その1つを可溶性画分より精製した。〔結果と考察〕ウシ副賢膜画分及び上清にはrho蛋白(Gb)のGTP水解活性を亢進する活性が見られ、その量比は4:6であった。このうち、細胞上清の活性を硫安分画のあと、phenyl Sepharose,CMーSepharose,TSKゲル、Mono Sの各クロマトで精製した。精製標品はSDSーPAGE上分子量28Kに単一バンドを呈した。精製倍率は約36,000倍、収率は0.6%であった。この蛋白は濃度依存的、時間依存的にrho蛋白(Gb)のGTP水解を亢進した。この亢進はrho蛋白(Gb)に特異的であり、ras蛋白のGTP水解には無効果であった。これらのことから、今回精製した蛋白はrho蛋白(Gb)に特異的なGAPであると結論された。Gbを初めとする低分子量GTP結合蛋白質は極めて低いGTP水解活性をもち、その活性の制御はGAPによって行われていると考えられている。今後、このGAPがどのような生理刺激で活性の制御を行っているかを検討する予定である。
