高等植物遺伝子のタッギングの為の基礎研究

研究概要	一定のDNA断片を高等植物染色体上の遺伝子内に挿入して不活性化させ、挿入DNA断片を不活性化された遺伝子と連鎖させる遺伝子タッギング法は未知の遺伝子の同定、マッピング、クロ-ン化、遺伝子発現調節機構の解明などのための有効な手段になり得ると考えられている。本研究では、プロモ-タ-を持たない薬剤耐性標識遺伝子を含むDNA断片や、トウモロコシの転移因子<Ac>___ー/<Ds>___ーをタバコヤイネのプロトプラストに直接法で導入し、形質転換体を得て植物個体を再生させてトランスジェニック植物を作出し、種子繁殖を行い、再生個体や次世代及び三世代植物中での導入DNA断片の安定性を検討して、高等植物タッギングのための基礎的デ-タを得ようとするものである。 (1)遺伝子タッギングにおいて、導入DNA断片と不活性化された遺伝子の両者が共に安定に次世代に伝達されることは重要な要件である。プロモ-タ-を持たぬ導入薬剤耐性標識遺伝子が植物遺伝子のエキソンに挿入された場合の安定性を検討するためのモデル実験として、ハイグロマイシン耐性(Hm^r)とカナマイシン耐性(Km^v)の両耐性遺伝子がbiーcistronic mRNAの生成により発現されるようなプラスミドを導入したトランスジェニックタバコを作出、次世代及び三世代への両耐性の伝達を検討したところ安定に伝達されていた。現在三世代目において希に起る不定性を検出できるか否かを検討中である。 (2)トウモロコシの転移因子<Ac>___ーや<Ds>___ーを遺伝子タッギングに利用するため、これらの転移因子が転移に伴って切り出されるとHm^r遺伝子が活性化されるようなプラスミドを導入し、Hm^r形質転換体よりトランスジェニックタバコやイネを作出、導入転移因子の転移を確認した。現在転移因子の体細胞における転移と生殖過程での転移を比較検討している。
