研究概要	糖質は細胞認識に関与するマ-カ-を始めする多くの重要な生物学的機能を果たしていることが明らかになってきたが、それらの生物活性糖質がその微細構造をどのように認識されて機能を発現しているかについては、まだ十分理解されていない。本研究では活性糖質の探索と構造の解析、合成研究にとどまらず、三次元構造の解析と生物活性の研究を組み合わせた広い視点から、糖質の構造認識と機能の発現機構を明らかにすることを目指して各研究者が下記の研究を行った。楠本は強力な免疫増強活性と毒性を示す天然糖脂質リポ多糖の活性部位リピドAに親水性糖鎖を結合させた誘導体を合成し、その高度な精製法を検討して純粋な合成化合物による生体成分との相互作用解析を可能にした。これとならんでリピドA部分の構造修飾によるリポ多糖成分の多彩な活性と構造の関係の解明を進めた。小川は代表的な赤血球糖タンパク質で、糖鎖を含めた全構造が解明されているグリコホリンのクラスタ-部を対象に取り上げて、NeuNAcーGalNAc二糖を含むシアロシルTnエピト-プ二量体をはじめて合成し、遺伝子に直接コ-ドされていないために常に不均一さをともなう天然糖ペプチドに代えて純粋な合成品を提供する道を拓いた。永井はホルモンによる細胞増殖と糖脂質の変化の関係を解析し、GlcNAcトランスフェラ-ゼなどの特定の糖転移酵素がホルモン刺激で活性化されることを明らかにした。目黒は1,6ーアンヒドロ糖の光ブロム化と重水素試薬還元によって糖の6位をキラル重水素化する画期的な方法を開発し、すべてのヘキソ-ス、ペント-スを標識した。さらにこれらを用いてオリゴ糖を合成し、NMRスペクトルによって(1ー6)結合のまわりの立体配座をはじめて正確に解析した。
