発生過程におけるグロビン遺伝子発現のスイッチング機構に関与する転写因子の解析

研究概要	グロビン遺伝子においては個体発生に伴い数種類の遺伝子の発現が順次切り変わるスイッチングの現象が知られている。この機構を分子レベルで明らかにするために、成体にもかかわらず胎児型のグロビン遺伝子の発現が高い例(HPFH)につき解析を行なった。その結果、γグロビン遺伝子の遠位CCAATbox内ー114位にCーT変異を認めた(CCAAT-CTAAT)。さらにこの塩基置換とHPFHの表現型との連鎖をPCRーdot blotにより家族のサンプルを用いて確認した。次にこの変異の核タンパクの結合への影響を5'フランキング領域の2つのCCAATボックスを有するフラグメントをプロ-ブとしてゲルシフトアッセイを行なったところ、赤芽球系細胞では3本のバンド(B1,B2,B3)が見られたが、非赤芽球系細胞では非特異的なバンドを除外すると2本のバンド(B1,B2)のみであった。変異プロ-ブではB1が見られなかった。各種コンペティションアッセイの結果B2はCP1,B3はNFE1であることがわかった。B1について、CP1とmutually exclusiveな関係にあるCDPの可能性を検討したが否定された。ゲルシフトアッセイやDNaseI footprintingの解析から、CP1の近位CCAATboxに対するアフィニティ-が遠位CCAATboxに対してよりも高いこと、またB1は両CCAATboxへのCP1の結合によるものであることがわかった。変異によりNFE1の結合には影響が見られなかった。以上の結果から正の転写制御因子として知られるCP1が遠位CCAATboxについては負の因子として働く可能性が考えられた。現在この変異ならびに2つのCCAATboxの機能面での解析を培養細胞やトランジェニックマウスを用いて行なっている。今後CP1の赤芽球の分化過程におけるプロフィ-ルをこの因子の遺伝子のクロ-ニングを行なうことにより明らかにして、スイッチング機構の解明を目指す予定である。
