研究概要	本研究では、多段階発がんのメカニズムを発がんプロモ-ション期とプログレッション期に焦点をしぼり、次の3つの研究目標のもとに研究を進めた。 1.皮膚での発現しているCキナ-ゼ分子種の同定: マウス皮膚nPKCηを分離した。nPKCηはカルシウム結合領域であるC2領域を欠き、C1(システイン・リッチ・レピ-トを含む調節領域)とC3(活性領域)からなるカルシウム非依存性Cキナ-ゼ(nPKC)の構造をもっていた。nPKCηは他のCキナ-ゼ分子種と異なり、肺と皮膚に強く発現していた。マウスCキナ-ゼの各分子種を新たに分離し、マウス皮膚で発現しているCキナ-ゼを系統的に検索した結果、nPKCηが皮膚の主要な分子種であることが判明した。 2.マウス皮膚のCキナ-ゼ標的タンパク質の同定: マウス皮膚および培養ケラチノサイトではp80、p70、p40、p34、p28の5種のタンパクがCキナ-ゼの標的タンパクであるこのうち、p70が核基質の一つ、ラミンBであることを同定した。ラミンBは酸性度の異なる二つのスポットB1、B2に分れるが、Cキナ-ゼでリン酸化されるのはB2であった。 3.発がんプロモ-ションに伴う遺伝子発現の変化: 発がんプロモ-タ-は各種の遺伝子の発現を誘導する。パピロ-マがんでは、そのうち、TPAによって遅れて誘導される二次反応性が恒常的に発現していた。Cキナ-ゼ遺伝子の中では、nPKCσががんで2倍高い発現がみられた。皮膚特異的Cキナ-ゼnPKCηの発現には変化がなかった。
