腫瘍抗原認識の分子免疫学的解析と担癌個体のサイトカインカスケ-ド是正の研究

研究概要	担癌状態ではヘルパ-T細胞(Th)活性、特にCD4^+Th機能が選択的に抑制される。このTh機能抑制の細胞制・分子機構を担癌宿生で産生される液性因子(免疫抑制因子)の観点から解析し、以下の成果を得た。 1.種々の腫瘍細胞の培養上清又は腹水中にtransfornuing growth factorーβ(TGFーβ)が多量に産生されていることを確認した。 2.腫瘍細胞培養上清又は腹水を種々の濃度でcytotoxic T lymphocyte(CTL)培養に加えたところ、CD4^+Thが関与するCTL応答が最も著明に抑制されることを明らかにした。この抑制はrILー2の添加によって阻止出来たのでCTL前駆細胞自身は影響を受けることなく、CD4^+Thのヘルパ-機能が抑制されることを証明した。 3.上記培養中に抗TGFーβ抗体を加えることで、腫瘍細胞培養上清又は腹水による抑制を阻止出来たこと、及びこれら腫瘍細胞由来sampleの替わりに、rTGーβを添加する事によっても同時のCTL応答の抑制を認めた事より、上記免疫抑制は腫瘍が産生するTGFーβによることが示された。 4.腫瘍細胞培養上清又はrTGFーβはCTL応答のみならずマクロファ-ジ活性化にも抑制効果をますことが認められた。即ち、CD4^+ThからのリンホカインであるIFNーrによりマクロファジ-が活性化され抗腫瘍効果を示すのであるが、上記上清又はrTGFーβはCD4^+ThのIFNーr産生を著明に抑制し、二次的にマクロファジによる抗腫瘍効果を抑制することが証明された。 5.以上TGFーβ又はTGFーβ含有物質(腫瘍細胞由来sample)によるCD4^+Th機能の抑制が種々の実験系で示されたが、この抑制はCD4^+Thそのものに対する抑制効果で、CD4^+Th活性化に必要とされる抗原提示細胞の機能不全を来たすことによる二次的な抑制でないことも証明された。
