研究概要

遺伝子組換え、細胞融合などのバイオテクノロジ-の基本をなす技術の生物学的、遺伝学的知見は急速に蓄積したが、これらの知見を工学的立場から解析し、実際の工業生産に応用する研究は大変遅れている。酵素、微生物または動植物細胞を固定化して利用するバイオリアクタ-の応用は今後ますます発展する分野であり、集中的かつ総合的な工学的研究が必須である。
本研究ではこれらの分野の新しい展開とそれらの技術の体系化を達成しようとしたものである。この総合研究の分担者はこの分野における工学的研究の主導的立場の研究者ばかりであるが、次のような四つのグル-プに分かれて新しい方向の探究と体系化をめざして平成2年度及び3年度に研究した。
1.動植物細胞用バイオリアクタ-の開発ーーー医薬品などを効率よく生産するため、固定化操作の簡便化、生産物の連続的分泌システム、老廃物の除去システムなどを開発した。
2.新規バイオリアクタ-の開発ーーー重要な酵素反応系に適したシステムの開発及び酵素の改良を行なった。
3.非水溶媒系酵素反応による物質生産ーーー非水溶媒系でしか進行しない酵素反応あるいは粉末状酵素によってアスパルテ-ムなどの有用物質の効率的生産システムを確立した。
4.膜分離操作を伴なうバイオリアクターーー最近発展の著しい膜分離操作と生化学反応とを組合わせて、乳酸や酢酸などの生化学物質の大量生産システムを開発した。
