研究概要

ダイズ根粒菌の宿主特異性については、Rj4遺伝子保有ダイズ品種に見られる特定菌株による根粒形成の抑制は菌培養の初期にのみ生成される未知物質に関係があり、その物質は培養を継続すると消失するために親和性が無くなること、また土壌から分離した根粒菌には二つの進化系統があり、菌体外多糖の組成、リゾビトキシンやインド-ル酢酸の生産、ヒドロゲナ-ゼ型の間に密接な関係のあることなどを明らかにした。莢切除実験によりシンク能の窒素固定に及ぼす影響の大きいことが示され、硝酸態窒素の抑制効果はダイズ、アズキ、インゲンの順に大きいが、これは宿主の窒素炭素代謝に対する硝酸吸収の影響を反映して、根粒の生長の抑制が大きいためであり、根粒中のレグヘモグロビンのニトロシル化にも関係がある。ダイズの硝酸吸収は温度、pHには大きく左右されるが、0.1mM以上の硝酸濃度には影響されない。共生特異的遺伝子を単離するためサブトラクション法によりノジュリン遺伝子のcDNAクロ-ニングを行い、20個以上のクロ-ンを得、その塩基配列を解析し、その内10個が未知の遺伝子であった。根粒の老化過程で生ずる遺伝子産物の出現が遮光や硝酸処理により変動するか否かをmRNAを比較することにより検討し、この遺伝子発現がage特異性である可能性を示した。クロバ-根粒菌の根粒形成遺伝子nod gene 14kbを組み込んだプラスミドpRtO32を移行させたツルマメ根粒菌によりクロバ-に根粒を形成させ、宿主域の拡大が可能であることを示した。根粒菌は非宿主植物の根毛にも付着し、nod遺伝子の発現を誘導する処理(フラボノイド処理など)を行えば、カ-リングも起きることを明らかにした。
