研究概要

ホルモンは生体制御機構の中で重要な役割を果たしており、その劇的効果は生物学の基礎的研究のみならず、医学の臨床面からも知られている。このようなホルモンの生理作用が古くから明らかにされていたのに較べ、その作用機構についてはようやく最近明らかにされようとしている。ここでは、昨年度に引続き、特に遺伝子発現調節ということに注目し、病態と関連させ、ホルモン作用機構を解明するために研究班を組織し、重点領域研究の申請の準備を行った。
1.<研究状況>___ーーグルココルチコイドとグルカゴンの協調作用を肝臓の糖新生および尿素合成酵素の遺伝子について解析した(市原、森、市山)。作用機序の不明瞭なインスリンについては、解糖系酵素の遺伝子の上流にインスリン応答配列が存在することが明らかとなり、その配列に結合する因子を究明しようとしている(田中)。甲状腺ホルモンおよび活性型ビタミンDによる遺伝子発現調節機構とステロイドホルモンの作用機序との類似性が明らかになりつつある(黒木、藤井、尾形)。抗脂血剤がペルオシゾ-ムの脂肪酸代謝酵素遺伝子群を誘導するのは、その遺伝子の上流に応答配列が存在し、ホルモンと同様の作用機序に基づくことが判明した(橋本)このように遺伝子にシス配列とトランス因子が関与していることが示されてきているが、一方ではDNAの存在様式、即ちクロマチン構造変化がホルモン作用発現に重要であることが示唆され(大場、市原)、新しい手法で研究を進展させたいと考えている。また、ステロイドホルモンをはじめいくつかのホルモンは単に一遺伝子の発現制御というレベルではなく、細胞増殖に制御に働いており、この機構についても解析をしている(松本、杉本)。
2.<研究発表と申請書作成準備>___ーー(1)まず、班員全員による研究発表を京都で行い、討論および今後の方向について活発な意見交換を行った。(2)その後、少人数の班会議を大阪および東京で数回開き、研究方針を立て、重点領域研究の申請書作成を行った。
