病原性ウイルスの進化機構の解明と進化方向の予測

研究概要	本研究の目的は、「病原性ウイルス遺伝子を用いて、その進化機構を解明し、未来進化の実証可能な予測を行なう」ことであった。本年度では、特にエイズウイルスとA型インフルエンザウイルスに注目して、これらのウイルス遺伝子の塩基配列デ-タを用いて、塩基置換パタ-ンの推定とそれに基づく進化方向の予測を行なった。エイズウイルスにおいては、ヒトエイズウイルスの系統樹作成と祖先ウイルスゲノムの塩基配列の推定を同時に行ない、AとG間の塩基置換が全塩基置換数の約50%を占めることを明らかにした。この生物学的要因としては、様々な可能性が指摘できるが、基本的にはエイズウイルスの複製酵素である逆転写酵素が有する複製エラ-の特性によるものと考えられる。この突然変異の特徴とも言える特質は、エイズウイルスの進化機構の解明や進化方向の予測を行なう上で、極めて重要な資料と言える。また、エイズウイルスの系統樹から、日本で感染した患者のウイルスが、どの地方のエイズウイルスと最も近縁であるかを解打し、日本のエイズ患者におけるウイルスの地理的起源を明らかにする手法、「エイズウイルスの「統統樹診断」とも言うべき方法を開発した。この新手法は、エイズウイルスの分子疫学において、非常に有用な方法となるのであろう。また、インフルエンザウイルスの塩基配列デ-タに関しても、同様な方法を用いて、動物インフルエンザウイルスとヒトインフルエンザウイルスの系統関係を明らかにした。また、インフルエンザウイルスの塩基置換パタ-ンを解析し、エイズウイルスのそれとは、かなり異なる可能性を示摘した。これらの研究成果は、インフルエンザウイルスの造化機構と進化方向の予測を行なう上で、非常に重要であると考えられる。
