ペプチドホルモン受容体調節機構のエイジングに関する研究

研究概要	ペプチドホルモン受容体調節機耕のエイジングに関する研究において,平成3年度に以下の成果を得た。 (1)マウスでわれわれが発見したFSH受容体のダウンレギュレ-ションが,マウス一般の現象であるか否かを確かめるため数系統で比較したところ,ICR系雄マウスにC57BL系と同様の現象が認められた。 (2)エイジングに併うマウス子宮のプロラクチン受容体量の変化を調べたところ,2か月齢の若いマウスに比べて,7か月齢の成熟マウスではその量が約1/2に減少していることが分った。しかし,生後1か月から同じ系統のマウスの下垂体を異所性移殖して高プロラクチン血症にしておくと,プロラクチン受容体量の減少は生じなしことも明らかになった。 (3)ラットのゴナドトロピン受容体遺伝子はGータンパク質連結受容体ファミリに属し,このファミリの遺伝子で保存性の高い膜通過部位のDNA配列を用いて,マウスの精巣cDNAライブラリからクロ-ニングを行った。しかし,陽性反応を示した3つのクロ-ンを選別し塩基配列を調べた結果,ラットのゴナドトロピン受容体との共通性は見られなかった。この結果は精巣にはゴナドトロピン受容体以外にもGータンパク質と連結して情報を細胞に伝える受容体が存在することを示唆する。現在ラットとヒトの受容体の中でゴナドトロピン受容体に特異的に保存性の高い部位のcDNAを用い,再クロ-ニングを行っている。 (4)ゴナドトロピン放出ホルモン(GnRH)の産生細胞である終神経細胞は,脊椎動物のペプチド作動性ニュ-ロンの神経修飾作用の研究モデルとして適していることが分った。熱帯魚ドワ-フグ-ラミ-の脳標本を用い,終神経GnRH細胞の細胞内記録と細胞分染色により,終神経細胞の自発放電は膜の内因性の性質に起因していること,神経突起の投射域に存在するGnRH受容体を持つ細胞に作用する可能性が分った。
