研究概要	線維芽細胞はマスト細胞の主要な増殖因子の1つstem cell factor(SCF)を生産する。SCFのレセプターはKITで、マウスではW遺伝子座に、ラットではWs遺伝子座にマップされている。マウスW遺伝子座には夛くの突然変異が存在するが、本年度は、W遺伝子座の突然変異のいくつかを分子レベルで性格ずけ、分子レベルの異常と変異マウスの症状に一致がみられるかどうかしらべた。W^<sh>突然変異遺伝子は、コーディング領域に異常がみられず、細胞種特異的に転写障害がみられたので、制御領域の突然変異と考えられた。W^<sh>/W^<sh>マウスはマスト細胞を欠損し、W^<sh>/W^<sh>マウスの骨髄細胞をIL-3存在下で培養して得た培養マスト細胞ではKITの転写はおこらない。W^<jic>とW^nはともに日本で発見されたW遺伝子座の突然変異であり、ともにKITのタイロシンカイネース領域の点突然変異である。ホモのW^<jic>/W^<jic>マウスとW^n/W^nマウスはともにマスト細胞を欠損しているが、ヘテロでは、W^<jic>/+マウスでマスト細胞が減少するのに、W^n/+マウスでは減少しないことがわかった。この原因はW^<jic>から生産されるKIT蛋白質が細胞外に発現されるのに対して、W^nから生産されるKIT蛋白質が細胞外に発現されないためである。KIT蛋白質は2量体を形成してレセプターになるが、細胞外に発現しないKIT蛋白質はレセプター形成に参画しないためである。W^<jic>/+マウスでは異常KIT蛋白質を含み、機能のないレセプターがつくられるのに対して、W^n/+マウスではレセプターの数は1/2になるものの、つくられるレセプター自身の機能は正常だからである。KITレセプターの機能をほぼ欠損するWs/Wsラットを用いて、マスト細胞同様、即時型アレルギー反応のエフェクター細胞である好塩基球の分化にはKITを介したシグナル伝達は必要ではないことを示す成績を得た。
