ILー5とその受容体系による免疫機能制御と炎症反応の調節

研究概要

ILー5はマウスB細胞増殖因子として同定され、その遺伝子が単離された。リコンビナントILー5はB細胞のみならず好酸球の増殖・分化を誘導するなど、多種類の細胞群に多彩な作用を示す。本研究は免疫機能制御と炎症反応におけるILー5とその受容体系の役割について、細胞レベル、遺伝子レベルで解析することをその研究目的とした。その結果以下の事を明らかに出来た。(1)マウス骨髄細胞の長期培養系において、ILー5は骨髄支持細胞とともに、Lyー1陽性早期B細胞及びLyー1陽性マクロファ-ジ前駆細胞の増殖因子と作用する。(2)粘膜免疫に主たる役割を果すと考えられるIgA産生に関し、ILー5はマウスB細胞によるIgA産生を促進するがμ鎖→α鎖へのクラス変換を誘導せず、すでにIgA産生を運命づけられたB細胞の成熟因子として作用する。(3)抗マウスILー5受容体抗体を用い、発現クロ-ニング法により、マウスILー5受容体cDNAの単離に初めて成功した。リコンビナントILー5受容体は分子量約60kDで、細胞外ドメイン、細胞膜貫通ドメイン、細胞質内ドメインより成り、低親和性ILー5結合能を示す。ILー5受容体はユニ-クなタンパク質であるが、細胞質外ドメインに他のサイトカイン受容体と共通のモチ-フを持っており、サイトカイン受容体ファミリ-に属することが、cDNAの塩基配列より推定されるアミノ酸の一次構造より明らかとなった。ILー5受容体cDNAをFDCーP1細胞(ILー5結合能を示さない)に遺伝子導入すると、機能的な高親和性ILー5受容体が検出できた。この事実は今回単離したILー5受容体と会合できる分子がFDCーP1細胞内に存在する事が示唆している。(4)マウスILー5cDNAを遺伝子導入して作成したILー5トランスジェニックマウスでは好酸球増多、血中IgA値の上昇、IgMクラスの自己抗体の産生が認められた。この事よりILー5は生体内でも免疫系免症系に重要な役割を示す事が確認された。
