ILー5とその受容体系による免疫機能制御と炎症反応の調節

研究概要

ILー5はマウスB細胞増殖分化因子として同定され、その遺伝子が単離され全構造が決定された。ILー5はB細胞のみならず好酸球の増殖・分化も誘導するなど多種類の細胞に多彩な作用を発揮させることも明らかとなっている。本研究は免疫機能制御と炎症反応におけるILー5とその受容体系の役割について細胞レベル、遺伝子レベルで解析することをその研究目的とした。その結果以下の事を明らかに出来た。(1)ILー5トランスジェニックマウスで増加した好酸球上にはB細胞上に発現しているものと同一のILー5受容体が発現しており、抗ILー5抗体や抗ILー5受容体抗体を投与すると好酸球増多は消失した。(2)健常人や好酸球増多症患者の血液中より好酸球を精製し、ILー5受容体の性状を検索した。ヒトILー5受容体は分子量約60kDで一種類の親和性を示すことがわかった。好酸球増多症患者の血中好酸球上のILー5受容体の性状は、健常人の好酸球上のそれと変りなかった。(3)寄生虫感染マウスに抗ILー5抗体、抗ILー5受容体抗体を投与し虫卵排除におけるILー5の役割を検索した。いづれの抗体も一次虫卵排除には影響を与えなかったが、2次感染における虫体排除を遅延させた。(4)マウスILー5受容体cDNAを遺伝子導入したL細胞,抗ILー5受容体抗体R52,抗ILー3受容体抗体(抗Aicー2)を用い、それ自身ではILー5結合能を示さないがILー5結合分子(α鎖)と会合し、機能的な高親和性ILー5受容体を構成する分子(ILー5受容体β鎖)を同定した。β鎖は分子量130kDでありGMーCSF受容体β鎖と同一であること、低親和性ILー3受容体α鎖と非常に類似している分子、ALC2Bであった。ILー5受容体β鎖はやはりサイトカインレセプタ-ファミリ-分子に共通に保存されている領域を保存していた。(5)胸腺内にILー5受容体β鎖を発現しているB細胞の存在を認識した。
