日本人I型糖尿病病因遺伝子の分子生物学的研究

研究概要	白人I型糖尿病発症において、HLAーDQB1遺伝子、特にその遺伝子によってコ-ドされるDQβ鎖57番目のアミノ酸の重要性が指摘されている。今年度は、日本人I型糖尿病発症に及ぼすDRB1、DQB1、DQA1遺伝子の影響について、DNAレベルで検討した。日本人I型糖尿病患者22名、および日本人健常人32名の末梢血リンパ球からDNAを抽出した後、DRB1、DQB1、DQA1遺伝子に特異的なプライマ-を使用したpolymerase chain reaction(PCR)を施行し、各遺伝子を増幅した。増幅したDRB1、DQB1遺伝子についてはallele specific oligonucleotideを用いたdot blot解析、DQA1遺伝子については制限酵素HaeIIIを用いたrestriction fragment length polymorphism解析を行い、各対立遺伝子およびDQ、DRβ鎖の57番目のアミノ酸を決定し、患者群および健常者群で比較した。その結果、1)白人I型糖尿病とは異なり、DQβ鎖57番目のアスパラギン酸は、日本人I型糖尿病の発症を抑制し得ないこと、2)DQw1.2は白人同様、日本人I型糖尿病発症を抑制すること、3)DRβ鎖57番目のアミノ酸は、日本人I型糖尿病のマ-カ-とならないこと、4)日本人I型糖尿病と密接に関係しているのはDQα鎖をコ-ドするDQA1遺伝子であり、α鎖のサブタイプのA1は発症抑制的に作用し、A3は発症促進的に作用していることが判明した。以上のことより、日本人I型糖尿病発症には、DQα鎖の構造が極めて重要である可能性が示された。
