研究概要

1.大腸菌繊維状ファ-ジのフロ-リングサ-クル型複製における複製型複開始機構について研究した結果、(1)複製エンハンサ-非依存性変異をもつ複製開始蛋白ではnicking活性対closing活性の比が極めて高く、複製開始の負の調節機構が欠損している、(2)エンハンサ-領域内でIHF蛋白に弱い結合を示す部位はそれ自身で湾曲構造をとり、大腸菌HーNS蛋白が特異的に結合する、(3)IHFとHーNSの二重変異株ではファ-ジ増殖がIHF単独異株に比べ1/10以下、野生型に比べ1/100以下に抑制される、(4)エンハンサ-領域内で複製方向とは逆向きの転写活性がin vitroでもin vivoでも認められること等を見出した。これら諸因子と複製調節の関係を今後検討して行きたい。2.同ファ-ジのマイナス鎖合成開始にあずかるRNAプライマ-の構造について解析したところ、これまで報告されていた場所より20塩基下流から合成されていることを見出した。3.大腸菌の熱ショック蛋白であるDnakの高温感受性変異株で染色体複製開始が高温で阻害される現象を解析した結果、複製開始蛋白DnaAの熱による不活性化が野生型Dnak蛋白によって著しく抑制されることを見出した(予研・榊原祥公氏と共同)。4.大腸菌の不飽和脂肪酸生合成遺伝子fabBの変異株においてオレイン酸補給による生育が高浸透圧条件により阻害されることを見出した。そこで浸透圧制御への関与が知られている転写因子OmpRと外膜脂肪酸受容体FadLの発現について調べた結果、(1)fabB株でOmpRを欠失させるとオレイン酸存在下低浸透圧でも生育できない、(2)fadLのrepressorであるfadRの欠失株によるオレイン酸の取込みは高浸透圧で阻害される、(3)fadLプロモ-タ-の上流及び下流のDNA領域には複製したリン酸化OmpRが特異的に結合することがわかった。これらの結果からFadLの発現はリン酸化OmpRを介した浸透圧制御を受けることが示唆される。
