研究概要

1.大腸菌繊維状ファージのマイナス鎖合成は、1本鎖DNA(プラス鎖)上の特定部位でRNAポリメラーゼに依るプライマーRNA合成によって開始される。(1)当該DNA領域の種々の変異を作り、origin機能に必要な領域を二つのヘアピン構造(B及びC)中に特定した。一部の領域では、二次構造のみならず特定の塩基配列を要することを明らかにした。(2)プライマー合成とDNA合成をcoupleさせたin vitro系において、RNA合成のみの系と同じ位置から同じ長さのRNAがプライマーとして合成され機能することを示した。(3)RNAポリメラーゼとoriginDNAとの結合をfootprintingにより解析し、RNAポリメラーゼはσ^<70>因子に依存してヘアピンBのstemの下半分程からヘアピンCのstemの下半分程までの、約50数塩基の範囲に結合することを示した。(4)従来プライマーRNA合成と転写開始反応は全く異なる反応と考えられてきたが、プライマーRNA合成開始点の決定(昨年度)及び上記footprintingの結果等から、両反応機構には共通性があるものと考えられるに至った。2.同ファージのプラス鎖複製開始機構については、(1)core領域へのgp2の結合はDNAを、complexIで90度、complexIIでは約160から180度(U字型)曲げること、複製エンハンサーにIHFが結合してDNAを約140度曲げることを電顕によって示した。(2)当該領域における種々の変異体の解析により(a)コア領域とエンハンサー領域間の距離が複製効率に影響しphasing効果が認められること、(b)エンハンサー領域内のIHF結合site1とsite2',2"の間にもphasing効果があること、(c)IHF結合siteをIHF蛋白の結合不能な配列に変えると複製効率が低下することを示した。3.大腸菌複製開始蛋白DnaAの熱不活性化がDnaK蛋白によって抑制される反応において、DnaAとDnaKは1対1の複合体を形成することを示した。熱抵抗性獲得もDnaKとの複合体形成も、共にATPの存在で阻害されるが、ATPの水解は必要としない。
