研究概要

1.大腸菌繊維状ファージのプラス鎖DNA複製は、負超〓旋をもつ環状二本鎖DNAにおいて、複製開始蛋白gpIIが特異的に単鎖切断を導入することによって開始される。前年度までの研究から、この時gpIIが段階的に結合してDNAを湾曲することを示したが、今年度はgpIIの結合によって、隣接する単鎖切断の起こるべき領域の二重鎖構造が一本鎖に開裂することを、過マンガン酸カリを用いる方法によって明らかにした。この開裂は、DNAが負の超〓旋をもつ場合にのみ認められた。また化学合成したDNAを用いることにより、直鎖状のDNAであっても、gpII結合領域が二本鎖で単鎖切断領域が一本鎖であれば、極めて能率よくgpIIによる切断が起こることを示した。切断領域が二本鎖であったり、結合領域が一本鎖であった場合には、切断は起こらなかった。以上から、二段階にわたるDNAの湾曲とそれに続く二本鎖構造の開裂が、複製開始反応に先立って起こらなければならないこと、負超〓旋が切断に必要なのは切断領域の二本鎖構造の開裂を起こすためであることが明らかになった。
同ファージのマイナス鎖複製は、一本鎖DNA上の特定部位をRNAポリメラーゼが認識してプライマーRNAを合成することによって開始される。このDNA部位は、2つのヘアピン構造を作ることによって部分的に二本鎖構造をとる。両ヘアピン部位に種々の変異を作成して同酵素との相互作用を検討した結果、プライマー合成反応と転写開始反応には共通の機構があることを強く示唆する結果を得た。なお、プライマー合成欠損変異ファージは感染した菌にSOS誘導を起こし、これはSOS誘導シグナルが一本鎖DNAであることを示唆することをさきに報告したが、その実験では低レベルのファージ増殖を伴うので、今年度は蛋白合成を完全に阻害した条件で感染を行い、SOSシグナルの出現をLexA蛋白の分解によって検出することにより、SOS誘導シグナルが一本鎖DNAであることを最終的に証明した。
