細菌感染防御機構におけるγ/δ型T細胞の意義

研究概要

γ/δ型T細胞の細菌感染防御機構における役割を検討するため、マウスを用いSalmonella choleraesuis感染症でのγ/δ型T細胞の動態、Vレパートアー、特異性ならびにその機能を解析した。BALB/cに致死量以下のS.choleraesuis RF-1(強毒株)または31N-1(弱毒株)を腹腔内投与すると、弱毒株で感染3日目及び6日目に腹腔内と肝臓内でγ/δ型T細胞が有意に増加した。一方、強毒株感染では菌の増加にもかかわらず、感染部位でのγ/δ型T細胞の増加を認めなかった。弱毒株感染で増加してくるγ/δ型T細胞は主にVγ1/Vδ6のV遺伝子を使用しており、CD4^-CD8^-CD44^+LFA-1α^+β^+IL-2Rα^-β^+であった。このγ/δ型T細胞はサルモネラ死菌とマイコバクテリア由来のPPDに反応して有意に増加した。強毒株感染マクロファージと弱毒株感染マクロファージに発現されるストレス蛋白質(HSP)の量をHSP65に対するmAb,IA10を用いてウェスタンブロット法で比較したところ、弱毒株感染マクロファージで強い内因性HSPの発現が認められた。さらに腹腔非附着性細胞をこれら感染マクロファージとin vitroで培養すると、弱毒株感染マクロファージと培養した時にγ/δ型T細胞の有意の増殖を認めた。γ/δ型T細胞の抗原認識にマクロファージ由来のHSPが重要と考えられる。次にサルモネラ感染の感染性を規定しているIty遺伝子のγ/δ型T細胞誘導における関与を検討するためにIty-resistant DBA/2,及びIhy-sensitive BALB/cでのS.choleraesuis31N-1(弱毒株)感染症におけるγ/δ型T細胞の出現の差を調べた。DBA/2感染マクロファージには強い内因性HSPが発現されており、γ/δ型T細胞の増加も著しかった。逆にBALB/cではHSPの発現が弱く、γ/δ型T細胞の増加率も小さいことがわかった。
以上の結果より、感染早期に出現してくるγ/δ型T細胞はマクロファージの発現する内因性HSPを認識して増加することが明らかとなった。
