研究概要	ヒト血小板を用いて、PLCの各種アイソザイムとGタンパク質を分離・精製し、機能的再構成系でそれらの相互作用を検索することを目的として研究を行った。ヒト血小板には数種類のPI-PLCアイソザイムやG蛋白質が存在しており、種々のカラムクロマトグラフィーにより分離・精製し、既知の各種アイソザイムの抗体を用いて同定を行なった。サイトゾル画分にはPLC-β(150kDa)、PLC-γ1(145kDa)PLC-γ2(145KDa)とPLC-δ(85kDa)が同定された。さらに未同定の活性画分の存在を確認した。また、膜画分には主にPLC-βが同定されたが、さらにmPLC-IIを精製した。一方、ヒト血小板のサイトゾルと膜画分より高分子量型(ヘテロトリマー)と低分子量型(モノマー)のG蛋白質を分離・精製し、それぞれの同定を行った。膜にはIAP感受性G蛋白質の主にGi2が同定され、他にGi3も少量含まれていた。またIAP非感受性Gタンパク質のGqの存在も抗体を用いて確認した。膜画分にはra1A、rac1、rap1Bが同定されているが、他にm22KGI、IIを精製した。また、サイトゾルからc21KG(smg21A、Krev-1)が多量に存在していることが確認され、他にc25KGを単離・精製し、部分アミノ酸配列から合成オリゴヌクレオチドを用いて、巨核球ライブラリーからcDNAをクローニングしramと命名。PLCの活性調節機構として、チロシンリン酸化について検討したが、トロンビン刺激によるPLC-γのチロシンリン酸化は確認されなかった。一方、トロンボキサンA2受容体にはGqがカップルしていることが明らかにされ、このGqはPLC-β1を特異的に活性化することが明らかになり、ヒト血小板におけるトロンボキサンA2-Gqα-PLC-β1の活性化経路が示唆された。また、ヒト血小板のPLC画分は細胞骨格系の主要構成成分であるアクチン-ゲルゾリン複合体と結合していることが示され、プロフィリン同様にPLC-γのゲルゾリンによる活性調節の可能性を示唆した。
