減数分裂期遺伝的組換えの分子機構

研究概要	減数分裂期には遺伝的組換えが体細胞分裂期の約1000倍の頻度で起こり、そのときにはシナプトネマ複合体や組換え節等の構造体が関与すると考えられている。我々はこの組換えの分子機構を明かにする目的で、体細胞分裂期の組換えは正常で減数分裂期の組換えのみが欠損した酵母S,cerevisiaeの変異株、MRE1(HOP1),MRE2,MRE3,MRE4/MEK1,MRE11を分離した。これらの変異株の解析の結果、MRE2,MRE3,とMRE11について次のことが明かになった。MRE2はRNA結合蛋白質に共通なアミノ酸配列を持つ蛋白質をコードしているが、この遺伝子が欠損すると、シナプトネマ複合体の形成が起こらず、対合しない染色体が観察される。そして第一減数分裂が野生株に比べて一時間早く始まるが、第二減数分裂の開始は両者ほぼ変わらない時間に開始する。この間、紡錘極体の分配の距離は、野生株の約2倍になる。このことは第一減数分裂時の微小管の伸張の長さからも確かめられた。従って、MRE2は染色体の対合と第一減数分裂の開始に働いている可能性がある。MRE3からは、体細胞分裂期と減数分裂期とでN末端の異なる2種類の蛋白質が発現していることが分かった。体細胞分裂期に発現している蛋白質は他方よりN末端が短く細胞増殖に関与している。欠損するとG1期が異常に長くなる。一方減数分裂期に発現する蛋白質が欠損すると、組換えが観察されずシナプトネマ複合体の前駆体であるaxial elementの形成も見られない。またMRE11はRAD50と極めて似た表現型を示すがRAD50の上位にあることが分かった。温度感受性のmre11ts変異株を用いた実験から、MRE11は組換えの初期過程、DNA二重鎖切断の挿入に関与し、切断後のprocessingの過程にも関与していることが分かった。
