研究概要

カテコ-ル体を分解する二原子酸素添加酵素には、水酸基の間を開裂するIntradiol型と水酸基の隣を開裂するExtradiol型がある。Extradiol型のカテコ-ル2,3ー二原子酸素添加酵素については既にDNA塩基配列を決定しているので、比較生化学的研究の一環として本年度はIntradiol型酵素の精製・クロ-ニングおよびExtradiol型酵素の活性部位の検索を試みた。
1)2,4ージクロロフェノキシ酢酸(2,4ーD)を唯一の炭素源として生やした緑膿菌より3,5ージクロロカテコ-ルの環開裂反応を触媒する酵素を均一にまで精製した。本酵素が3,5ージクロロカテコ-ルの2つの水酸基の間を開裂して2,4ージクロロムコン酸を生ずる反応を触媒するIntradiol型酵素であることを明らかにし、3,5ージクロロカテコ-ル1,2ー二原子酸素添加酵素と名付けた。基質特異性、反応動力学的定数、Nー末端アミノ酸配列の決定から、本酵素は従来報告されているクロロカテコ-ル二原子酸素添加酵素とは異なる新しい酵素であることを明らかにした。さらに、本酵素遺伝子がプラスミド(84kb)に存在することを明らかにし、本酵素遺伝子を含む1.6kbの制限酵素フラグメントのクロ-ニングに成功した。現在、本酵素遺伝子の塩基配列決定を進行中である。
2)カテコ-ル2,3ー二原子酸素添加酵素について、οーニトロフェノ-ルが有用な活性部位検索試薬となることを昨年度報告したが、本試薬を用いた動力学的解析より、活性部位の二価鉄と基質・酸素との結合様式について追究し,さらに,活性中心の鉄の他金属による置換実験、ならびにSH試薬や自殺基質による酸素の修飾を行い、活性に関与するアミノ酸残基の推定を試みた。
