細胞質分裂の分子機構に関する研究

研究概要	1)収縮環が細胞分裂のどの時期にどのような時間経過で形成されるかをウニ卵を用い、アクチン繊維の蛍光ファロイジン染色によって詳細に調べた。収縮環のアクチン繊維は核分裂終期に染色体胞が融合する時に初めて出現し、初めは表層の網目構造と構造的に区別できないが分裂が進むにつれて束ねられ、分裂溝が最大速度で進行する時には高密度の束になった。これらの過程に2分を要し、収縮環は瞬時に形成されるという通説は正しくないことがわかった。2)収縮環形成にミオシンがなんらかの役割を演じているとの作業仮説をたて、各種蛋白質リン酸化酵素の阻害剤がウニ卵の分裂におよぼす影響を調べた。ミオシンのリン酸化酵素の阻害剤MLー9をウニ卵に与えるとある濃度範囲では核分裂は起こるが細胞質分裂が阻害された。この時収縮環が形成されたかどうかを蛍光ファロイジン染色で調べたところ、形成されていなかったのでミオシンのリン酸化が収縮環形成に働いている可能性が示唆された。3)アクチン調節蛋白質プロフィリンは収縮環形成と消滅に関与していると考えられる。この蛋白質の機能を明らかにする一環として、ムラサキウニ卵、タコノマクラ卵、粘菌変形体のプロフィリンのアミノ酸配列を蛋白質化学的手法で決定した。これまでに知られているプロフィリンの配列と比べると、脊椎動物ーウニー粘菌間では相同性は低いが、N端部分とC端部分の配列は生物種を越えて保存され、これらの部位がアクチン結合部位であろうと考えられた。4)タコノマクラ卵第一分裂の分裂溝を界面活性剤を含む溶液を用いて初めて単離した。この画分には他の表層部分には見られない蛋白質が数種見られ、分裂に必要な成分であろうと考えられた。現在これらの蛋白質の同定を進めている。
