酸素感受性酵素の精製・活性測定装置の試作

研究概要	1.高度酸素感受性酵素の精製・活性測定装置の試作種々の因難はあったが本装置の試作に成功し、高度の嫌気条件を保ちながら、ここに、酵素活性測定用の分光々度計、酵素精製用のシステムなどを組み込むことができた。本装置の断熱効果もよく、酵素精製のための冷蔵室の温度も低く維持することができ、酵素活性測定室の異常高温を生ずることもなく、予期以上の良好な状態で運転することができた。 2.本装置を使用して得られた新たな知見 (1)嫌気的・低pHでの歯垢微生物の糖代謝上記なような条件で口腔レンサ球菌より多くの乳酸をつくる生化学的機構を明らかにすることができた。すなわち、低いpH条件下では、乳酸の酸性を触媒する乳酸脱水素酵素の生合成が促進され、糖制限・嫌気条件下で環境のpHが低下したときのレンサ球菌菌体内のpHを正確に測定することに成功した。低いpH条件下では細菌菌体内のpHもかなり低下し、至適pHの低い乳酸脱水素酵素が、至適pHの高いピルビン酸ギ酸リア-ゼ(PFL:ギ酸等の産生のキイ酵素)よりもはるかに活性が高くなることによって、低いpHで乳酸が多くつくられるようになることが判明した。 (2)高度嫌気条件下でのアクチノミセスの糖代謝この様な条件下でアクチノミセスが糖を代謝する際、重炭酸を添加すると糖代謝が著しく促進され、これが、菌体内のNADHのレベルの変換によっていることが明らかにされた。このことは、歯垢中に多く生息しながら、その酸産生能(糖代謝活性)が低く根面ウ蝕への関与があまり重視されていなかった本菌が、実際の口腔内のように、高度嫌気で、唾液よりの重炭酸の供給の多い条件下ではかなりよく酸をつくることを示し、これまで考えられていた以上に根面ウ蝕への重要な関与を示唆した。
