老人性神経細胞死を阻止するグルタミン酸遮断薬の設計と開発

研究概要

カイニン酸とアクロメリン酸の作用を比較する1つの方法として、cーfos mRNA発現の脳内分布を検討した。カイニン酸を全身投与した場合、海馬錘体細胞、歯状回顆粒細胞、扁桃核、梨状核にcーfos mRNAが発現したのに対し、アクロメリン酸の場合は、歯状回顆粒細胞のみであった。一方、脊髄においては、両者とも運動神経細胞に発現したが、アクロメリン酸の方がカイニン酸よりも顕著に発現した。cーfos発現の意味ずけはまだ全て明らかにされているわけではないが、神経細胞死とcーfos発現との関係について今後の発展が期待されよう。新生ラット脊髄において、アクロメリン酸よりも更に強力な興奮作用を持つ類似化合物を合成された。これはメトキシフェニル基を持つカイノイドで、構造的にアクロメリン酸に近いが、ラットに投与すると、これによって生ずる神経学的症状はカイニン酸とアクロメリン酸の両方の性質を呈し、カイニン酸受容体に2つのサブタイプが存在するとの我々の仮説をチェックするのに都合のよい物質である。この物質を用いて、カイニン酸受容体には生理学的に区別できる少なくとも2つ以上のサブタイプがあるのだろうと結論された。
現在は局所的に興奮性アミノ酸を適用して神経細胞死を起こさせる方法がかなりpopularになったが、神経細胞だけの片側半球壊死を起こさせることは技術的に困難であり、全脳虚血による神経細胞死の場合は、死亡率が著しく高い。私どもは、興奮性アミノ酸の特殊動脈内注射により、死亡率がほとんどゼロで片側半球壊死を起こさせることに成功した。この標本は、いろいろな角度から行なわれる神経細胞死の研究に大きく貢献することが期待される。次に脊髄運動神経細胞だけに損傷を起こす方法を開発中である。これは筋萎縮性側索硬化症のモデルを意識している。この他神経細胞死を防止する化合物を検索中である。
