前立腺由来の骨形成促進因子の同定およびその骨粗鬆症治療への応用

研究概要

前立腺癌や前立腺肥大症の前立腺組織中には、強力な骨形成促進活性が存在する。そこで、この前立腺由来の骨形成促進因子を同定すると共にその作用を解明し、ひいては老人性骨粗鬆症患者の治療への応用をはかることを目的として以下の検討を進めた。
1.骨形成促進因子の精製:前立腺肥大症患者より採取した前立腺組織の尿素抽出物をイオン交換および逆相HPLCにより粗精製し、三つの骨形成促進活性および線維芽細胞のprogression活性を有するピ-クを得た。このうちピ-ク1の精製を、progression活性を指標としてFPLCを用いたゲル濾過カラムなどにより進めた。そして、SDSPAGE上の主バンドアミノ酸部分構造を決定した。その結果、検討した中間部6個およびC端部6個、計12個全てのアミノ酸配列がangiogeninのそれと一致した。更に、ピ-ク2および3についても同様に精製を進めたが、充分な蛋白量が得られないこと、精製を進めることにより失活することなどより、その同定には至っていない。
2.骨形成活性の評価:Angiogeninの骨形成および骨芽細胞に対する増殖促進活性をin vitroで検討した結果からは、これらの活性は殆ど認められなかった。一方、前立腺抽出物中には、異所性骨化を指標としたin vivoでの検討により、骨誘導活性の存在が認められた。そこで更に、前立腺抽出物中の骨誘導活性に関する評価をin vivoおよびin vitroの両面から進めると同時に、強力な骨誘導活性を有する骨基質中の成長因子であるbone morphogenetic proteinとの異同について、FPLC上の溶出パタ-ンの比較およびWestern blot解析などにより、検討を進めている。
