前立腺由来の骨形成因子の同定およびその骨粗鬆症治療への応用

研究概要	前立腺組織中には多数の成長因子が存在し、前立腺癌の骨転移により骨形成性変化がもたらされる。そこで、前立腺由来の骨形成作用を示す成長因子を同定すると共にその作用を解明し、ひいては老人性骨粗鬆症患者の治療への応用をはかることを目的として以下の検討を進めた。前立腺肥大症患者の手術時に得られた摘出前立腺組織の尿素抽出物中にはcompetence成長因子活性とprogression成長因子活性の両者が存在したが、このうちprogression活性を示す分画中に強い骨形成促進活性が存在した。そこで、progression活性を陰イオン交換、逆相、ゲル濾過、陽イオン交換、逆相、ゲル濾過、heparin親和性、陰イオン交換そして逆相の各HPLCで精製した。以上の精製を経てもprogression活性はなお単一蛋白とはならなかったが、活性画分を採取し骨芽細胞様細胞株ROS17/2.8の増殖に及ぼす効果を検討したところ10nM IGP-1を上回る増殖促進活性を示した。しかしalkaline phosphatase活性の上昇作用は認められず、collagenベレットと共にラット皮下に移植後の異所性骨誘導作用も認められなかった。しかしこの因子の骨形成促進作用の有無はこれらの検討のみでは明確にすることはできないことから、今後in vivoでの骨膜下への注入などによりその骨形成作用を更に詳細に検討する必要がある。一方、この因子の同定をはかるため、得られた画分の酵素処理後peptideマッピングを行い各フラグメントのアミノ酸配列を同定した。その結果high mobility group protein-1、cysteine rich protein、histone H1BおよびH10などの核蛋白との共通配列が認められたが,既知の成長因子との相同性を示す配列は認められなかった。そこで骨芽細胞の増殖促進作用を示す新たな成長因子が存在する可能性を明らかにするため、前立腺細胞のcDNA libraryを用いこれらの蛋白のcloningを進めている。
