研究概要	本年度は生体試料として基本的な核酸とタンパク質について、無機材料研究用走査トンネル顕微鏡を用いて超高真空下で観察し、生体試料観察用走査型トンネル顕微鏡に必要な性能と操作性を最終的にまとめて、表記の目的で設計・製作した。生体試料の観察には生端形状ができるだけ鋭いタングステンの電解研摩探針を使うのが最良と判断し、この探針が空気中の酸素により酸化をうけ劣化しないよう、密封室内にトンネル顕微鏡をおき、試料及び基盤の出し入れは室内の状態を乱さずできるようにした上で、密封室内を清浄なチッソガスあるいはアルゴンガスで満たせるようにした。探針のとりつけ、基盤への試料の添加についても、手袋を通して操作できるようにし、清浄ガス中で行うこととした。この結果、空気中での油滴の沈着など有機物による汚れも大巾に減少した。超高真空型のトンネル顕微鏡でDNAとタンパク質の像を得る事に成功したので、現在はその解像度の向上に努めるとともに、真空でない、自然な湿度を保った状態での生体分子の観察をするために新しく試作したトンネル顕微鏡で条件を探索しているところである。DNAは、クラファイト基盤上で細状分子としてあるいは球状にかたまった分子として割合高い頻度で見る事ができ、よい条件下ではらせん構造を見分ける事も可能である。ただ探針の先鋭度がまだ十分でないため、十分満足できる解像度はないし、球状にかたまる性質を試料調整に際してなおして細状にのびたイメ-ジを得る努力が必要である。タンパク質については現在超高真空状態又は大気圧において映像を得る事がDNAより難しく、乾燥による変形も著しいようである。この点で今年度試作した機器の性能が十分引き出されるようになれば、清浄で適正な湿度下における変形の少ない生体分子の映像が得られる事を期待して実験を進めている。本年度は以上のように試料変性を最小限に止める生体観察法の開発途上にある。
