ビタミンE関連新規化合物の合成と生体における酸素毒性防御機構の解明

研究概要	1.ユビキノ-ルの活性ラジカル消去作用とビタミンE再生作用ユビキノ-ル(還元型ユビキノン)の活性酸素ラジカル消去速度K_1および、ビタミンE再生速度K_2をストップドフロ-分光光度計を用いて測定した。K_1およびK_2の値はそれぞれビタミンEおよびビタミンCに対して報告された値と同じオ-ダ-の値を示した。各種の生体膜中におけるビタミンEとユビキノ-ルの存在濃度を考慮すると、少くともミトコンドリア膜中ではユビキノ-ルが脂質の過酸化抑制に大きく寄与していることが明らかとなった。リポタンパク中でのユビキノ-ルの抗酸化作用への寄与は小さいが、肝臓・心臓など各種組織の膜中ではユビキノ-ルの寄与が充分期待できる。 2.エストロゲンの抗酸化作用とビタミンE再生作用女性ホルモン中でフエノ-ル骨格を有するエストロゲン類について、その抗酸化活性の速度論的評価を行った。エストロン、エストラジオ-ルなどの抗酸化活性はBHTなどの代表的抗酸化剤と同程度の活性しか示さない。一方、カテユ-ルエストロゲンはビタミンEよりも高い活性を示した。この物質が女性だけの体内で作られることを考えると、女性の長寿との関連が期待される。 3.ビタミンEの酸化促進作用の速度論的評価ビタミンEが高濃度で食用油脂中などに存在すると、逆に油脂の過酸化を促進することが知られている。この機構を明らかにするために、ビタミンEラジカルと不飽和脂肪酸類およびそのヒドロペルオキシド体との反応を行い、速度定数K_3およびK_4を求めた。K_3,K_4の値の比較から、ビタミンEラジカルと脂肪酸との間の水素引き抜き反応が酸化促進反応に寄与することを明らかにした。
