心房性ナトリウム利尿ホルモンの分泌様式における生物発生原則の評価

研究概要

1.ウナギの心室より、新しいナトリウケ利尿ペプチド、心室性ナトリウム利尿ペプチド(VNP)を単離し、その一次構造を決定した。このホルモンは、これまでに見つけられた3建類のナトリウム利尿ペプチド(心房性ナトリウム利尿ペプチド、ANP;脳性ナトリウム利尿ペプチド、BNP;Cーtypeナトリウム利尿ペプチド、CNP)と比較して、Cー末端が長く延びているところに特徴がある。
2.ウナギの心室からVNPのcDNPクロ-ニングして、その塩基配列を決定した。アミノ酸配列と同様に、塩基配列からみてもVNPはこれまでに見つかっていない全く新しいホルモンであることがわかった。
3.ウナギANPとVNPのラジオイムノアッセイ系をかくりつしてウナギの心房、心室、及び血漿中の濃度を測定したところ、心房ではANPがVNPの100倍、心室ではVNPがANPの5倍、血漿中ではVNPがANPの3倍含まれていた。心臓に貯えられている全量ではANPがVNPの100倍に達するので、VNPは心室で作られほとんど貯えられることなく血液中に分泌されていると予想される。
4.ウナギの心筋を細胞培養したところ、心房細胞ではANPは大量に貯えられていたが、心室細胞ではVNPはほとんど貯えられておらず、大量に培地中に分泌されていることがわかった。従って、心房では主にANPが合成されregulatory pathwayで分泌されているが、心室では主にVNPが合成され、constitutive pathwayにより分泌されているらしい。
