ATP依存性輸送膜蛋白質の機能の解明

研究概要

真核生物のATP依存性輸送膜蛋白質は、ATP加水分解のエネルギ-に依存して、ホルモン、薬剤などを輸送するポンプであり、癌の多剤耐性、細胞分化、遺伝病など生命の重要な局面で機能している。本研究は、癌の多剤耐性に関与するヒトPー糖蛋白質をモデルに用い、遺伝子工学的に改変した種々の変異体の活性を検討することによって、ATP依存性輸送膜蛋白質の作用機構を解明することを目的とした。
Pー糖蛋白質は2つのATP結合領域を持つが、それぞれの領域がATP加水分解および薬剤輸送にどのように使われているのかは不明であった。そこで全長および前半後半それぞれ半分をβーガラクトシダ-ゼ(βーgal)との融合蛋白質としてマウス培養細胞内で発現させ、抗βーgal抗体を用いて部分精製し、リポソ-ムに再構成した。Pー糖蛋白質はATPase活性を示し、細胞を多剤耐性にした。Pー糖蛋白質の前半半分も同様にATPase活性を示したが、細胞を薬剤耐性にしなかった。以上の結果から、前半半分が単独でATPを加水分解できること、しかし薬剤を輸送するには後半半分も必要であることが明かとなった。次にPー糖蛋白質のATP加水分解反応を速度論的に解析するため、Pー糖タンパク質の細胞内ドメインを膜結合領域から切り離し、大腸菌内で大量発現させ、精製した。その結果、後半半分の細胞内ドメインも前半半分と同様にATPase活性を示すことが明らかとなり、Pー糖タンパク質の2つのATP結合領域は独立的にATP分解機能を持つことが明かとなった。さらに遺伝病嚢胞性繊維症でCFTR遺伝子に見つかっている代表的な変異を導入しATPase活性を測定した結果、変異周辺の領域がATP加水分解に直接影響するのではなく、ATPのエネルギ-によって蛋白質が構造変化する過程に関与していることが示唆された。
