レセプタ-からみた脳内各種ニュ-ロン系相互の統合機構に関する研究

研究概要

前年度のうちに、脳内の各種レセプタ-分布についての、マクロ・オ-トラジオグラフィ-法による検索法を確立することができた。そこで、それらの方法を応用して以下の実験を行なった。
脳内コリン系ニュ-ロンの代表的な例として、Meynert核やdiagonal bandからの大脳皮質に投射する系を利用した。すなわち、イボテン酸をこの起始核に注入して、この系のコリン作動性ニュ-ロン群を選括的に障害して、痴呆モデル動物を作成した。これらのモデルラットについて、脳内レセプタ-の動態を検索した。その結果、脳内の特に大脳皮質部位でムスカリン性コリンレセプタ-が特異的に減少していることが明らかになった。
また、脳梗塞モデル動物として、中大脳動脈起始部を閉塞して、線条体外側部虚血梗塞モデル動物を作成し、水迷路試験による行動学的検索や脳内レセプタ-をマクロ・オ-トラジオグラフィ-法を利用して検索した。具体的には、右側の中大脳動脈を1時間虚血の状態にした。その後、再開通させ、3週間後と3箇月後についてレセプタ-・オ-トラジオグラフィ-法で検索した。また、この様に作成した線条体外側部虚血梗塞モデルラットについて、空間認知をとらえるとされている水迷路試験での動物の行動を詳細に観察した。水迷路試験で、虚血群は対照群にくらべ、有意にプラットホ-ムへの到達時間の延長をみた。すなわち、虚血により水迷路試験での学習効率の低下が認められた。レセプタ-・オ-トラジオグラフィ-法ではGABA系を中心に観察した。アゴニストであるムチモ-ルによるオ-トラジオグラムではGABAレセプタ-が梗塞部位に一致して減少していた。また、GABA Aのアンタゴニストであり、より特異的なリガンドとしてホモジュネ-ト結合実験で使われているリガンド(SR 95531)を利用したオ-トラジオグラムでは、ムチモ-ルのものよりも明瞭な減少が認められた。
