松果体に存在するLH-RH様物質の同定及びその生物学的役割に関する研究

研究概要

松果体に存在するLH-RH(GnRH)様物質及びその生物学的役割に関する研究に於て平成4年度に以下の成果を得た。
52kDタンパク質の生化学的性質を詳しく調べるため、2次元電気泳動を行った後、イムノブロットで検出した。その結果二つの異なる等電点を持つ蛋白質からLRH13との免疫反応が得られ、52kD蛋白質は2種類の異なる等電点、即ち6.8と7.0を持つ蛋白質の集合であることが分かった。また蛋白量としては、等電点6.8のものが主であったが、単クローン抗体LRH13との反応性には大きな差は見られなかった。
これら2種の蛋白質が異なる等電点を持つ理由を調べるために、V8蛋白質分解酵素を用いて部分分解を行い、ペプチド・マッピングを行った。その結果、双方の蛋白質は同じパターンで部分分解され、同一のアミノ酸配列を持っているものと考えられ、等電点の違いは、mRNAからの翻訳後に起こった修飾の差に起因している可能性が生まれた。
52kD蛋白質が二つの等電点を持つ理由の一つとして糖鎖修飾の違う可能性を検討するため、糖鎖に結合するconcanavalin Aとwheat germ agglutininを用いてレクチンブロットを行った。SDS-PAGE後のブロットで両レクチン共に52kD周辺の蛋白質と反応することが分かり、現在2次元電気泳動と組み合わせたレクチンブロットを検討している。
以上の結果から、GnRH様52kD蛋白質は同じアミノ酸の配列に異なる糖鎖の付いた糖蛋白質であり、それが等電点の違いをもたらしている事が判明した。現在蛋白質分解酵素で得られた各断片とLRH13およびレクチンとの反応性を調べており、それらのアミノ酸配列をもとにラット松果体のLHRH様蛋白質の本体を明らかにして行きたい。
