神経活性ペプチド前駆体遺伝子発現の薬物による調節ーNeuropeptide Y及びEnkephalin前駆体についてー

研究概要

神経特異的ペプチドNeuropeptide Y(NRY)及びEnkephalin A(ENK)は神経系(神経細胞)に最も豊富かつ広汎に発現し、そのプロセッシング産物はCotransmitter/neuromodulatorとしてシナプス伝達の調節に極めて重要である。これらの神経ペプチドのシナプス伝達の調節は生理的に遺伝子発現が変化し、そのペプチド量が変化する事で変動を受ける事を明らかにした。この遺伝子発現変化は神経活動によって修飾される点で、神経細胞のシナプス可塑性のメカニズムの解明に役立つ事も明らかにした。神経作働薬(降圧剤、アドレナリン作働薬)は、神経活動(transsynaptic)や膜受容体を介して、これらペプチド遺伝子の発現を転写レベルで変化させ、シナプス伝達の調節をおこす事も報告した。以上、これらのペプチド遺伝子発現の研究が、薬理学研究の領域拡大に役立つ事が明らかになった。
神経活動に伴うENK,NPY遺伝子発現の変化は細胞内2ndメッセンジャ-として膜依存性Caチャンネルを介する外液Caを使う事を示した。Nuclear runーon assay及びChloramphenicol acetyltransferase(CAT)assayを用いてこの転写活性増加に関与するCaーresponse element(CaーRE)を同定した。NRY遺伝子発現に関しては、細胞内2ndメッセンジャ-cAMPによるNPY遺伝子発現の活性化、糖質コルチコイドステロイドホルモンによる活性化、神経成長因子NGFによる神経分化に伴う活性化を報告した。これらの遺伝子発現活性化が、Nuclear runーonやCAT assayによって転写レベルで生じる事とcAMP response element,Glucocorticoid response element,NGF response elementを同定した。以上、分子生物学的手法で、神経遺伝子発現レベルにおける神経作働薬の作用機序を解明しつつあると考えている。
