研究概要

1.β2‐ミクログロブリン(β2‐M)とAmyloid Enhancing Factor(AEF):長期血液透析患者でβ2‐M由来のアミロイド(Aβ2M)が沈着した骨髄からAxerlad法により抽出した物質をマウスに注射したが、アミロイドの沈着を認めずAEFの活性は証明出来なかった。AEFはAA,ALアミロイドの沈着臓器やアルツハイマー病患者脳には存在すると報告されているが、透析アミロイドーシスにはAEFが関与しないか、沈着したAβ2Mが微量のために、発症を促進するに十分な量のAEFが含まれていなかったか、更に検討する必要がある。
2.AEFとユビキチンの異同について:最近、ユビキチンはAEFそのものであるとAli‐khanらが報告している。この報告を確認するために市販の抗ユビキチン抗体を使用して免疫組織化学的検討を行ったが、マウスの実験的アミロイドーシスではアミロイド沈着部位には陽性反応は認められなかった。また市販のユビキチン(Sigma)をマウスに静注(1mg/匹)し、10%カゼイン溶液0.5mlを3回皮下注射したところ、アミロイドの沈着を認めた。これはユビキチンにAEF活性がある事を示唆するが、アミロイドは脾濾胞周辺帯に僅かに見られるにすぎずAEFとしての活性は弱いと考えた。また、抗ユビキチン抗体を結合させた免疫アフィニティーカラムにAxerlad法により抽出したマウスの粗製AEFを通して得た抽出物を注射したマウス、吸着されなかった成分を注射したマウスのいずれにもアミロイドの沈着を認めた。この所見からはユビキチンがAEF活性があることは認めるが、ユビキチン=AEFとは言えないと考えた。1,2.の実験は少数の動物を使ったもので、今後、再実験を行いより明確なものにしたい。
