腸炎ビブリオの耐熱性溶血毒の病原性に関する研究

研究概要

腸炎ビブリオおよび類似ビブリオの産生する耐熱性溶血毒(TDH)の病原的役割を明らかにする為に、以下のような順序で研究を進め、それぞれに示したような成果を得た。1.腸炎ビブリオはTDHのみならず、類似する溶血毒(TDHーrelated hemolysin,TRH)も同時に産生する可能性があるので、TRHを特異的に検出するDNAプロ-ブを用いて、TRHを産生する能力のある菌株とない菌株を区別した。2.患者から分離された神奈川現象陽性分離菌株は、TDHをコ-ドする遺伝子を2コピ-持つ(tdh1およびtdh2)。その代表菌株として、TRH産生能力のないAQ3815株を選出した。3.AQ3815株のtdh1遺伝子をin vivo recombination法によって変異遺伝子置換して、不活化したところ、殆ど溶血活性の低下が認められなかった。従ってtdh1遺伝子は殆ど発現されず、むしろtdh2遺伝子が菌の溶血活性を担っていると考えられた。4.AQ3815株のtdh2遺伝子のみをsuicide vectorを利用して不活化したところ、溶血活性は殆ど無くなったが、感度の高いELISA法でTDHの産生を測定したところ、残存するtdh1遺伝子の発現によって、野生型菌の産生するTDHの約5%に相当するTDHが産生されていることが明らかになった。5.AQ3815株の2コピ-のtdh遺伝子の両者の不活化し、TDHをまったく産生しないisogenic mutantを作製した。この変異株と野生型AQ3815の培養上清を、ウサギの腸管粘膜上皮をUssing Chamberにマウントた実験系で比較した。その結果、培養上清中のTDHは粘膜上皮のイオンの透過性を変化させており、腸管内の分泌作用に影響を及ぼしている可能性が示唆された。
