アクロメリン酸の選択的神経細胞障害の発現機序:グルタミン酸受容体との関連

研究概要

アクロメリン酸は特異な選択的神経細胞障害作用を持つ非NMDAタイプの強力な興奮性アミノ酸であるが、その作用する受容体を明らかにする目的でグルタミン酸受容体に対する結合能を測定した。1.[ ^3H]カイニン酸、[ ^3H]AMPAのラット脊髄シナプトゾ-ム分画に対する結合能をフィルタ-法により測定し脊髄におけるカイニン酸、AMPA受容体の特性を明らかにした。脊髄のカイニン酸受容体、AMPA受容体とも結合部位は一種類であり、それぞれの解離定数(Kd)とBmaxは6.2+0.9nMと52+4.0fmol/mg protein、および7.6+1.5nMと70+5.6fmol/mg proteinであり、共に脳でのそれぞれの受容体の高親和性結合部位に近い性質を持つことが明らかになった。2.これら受容体に対するアクロメリン酸を含む種々のリガンドの結合能の強さを[ ^3H]カイニン酸(2.2nM)、[ ^3H]AMPA(3.0nM)結合の阻害作用により測定した。親和性の高さは、カイニン酸受容体ではド-モイ酸>カイニン酸>アクロメリン酸>>キスカル酸=AMPA、AMPA受容体ではキスカル酸>AMPA>アクロメリン酸>ド-モイ酸>カイニン酸の順であった。アクロメリン酸のAMPA受容体に対する親和性がカイニン酸より強いが、比較的純粋なカイニン酸受容体を持つ脊髄後根神経C線維においては、AMPA、キスカル酸共に弱い脱分極作用を持つにもかかわらずアクロメリン酸がカイニン酸よりも高い脱分極作用を持つのでAMPA受容体に働くとは考えにくい。これらの結果はアクロメリン酸の極めて強力な脱分極作用および選択的神経細胞毒性の発現には、カイニン酸受容体、AMPA受容体のいずれとも異なった受容体(アクロメリン酸受容体)が関与しているることを強く示唆する。3.Autoradiographyは現在進行中である。
