血管平滑筋におけるrasP21類似低分子量GTP結合蛋白質の役割とその病態

研究概要	ras癌遺伝子の産物(ras p21)は分子量約2.1万のGTP結合蛋白質(G蛋白質)で細胞増殖の制御に関与するが、最近、ras p21のGTPase活性促進蛋白質(ras p21GAP)がPDGFの作用時にチロシンリン酸化されることが明らかにされ、ras p21がPDGFの情報伝達機構に関与することが決定的になっている。私共は血管平滑筋におけるras p21類似G蛋白質について以下の成果をあげている。(1)血管平滑筋細胞にはras p21と共にras p21の作用に拮抗するras p21類似G蛋白質のsmg p21が多量に存在するが、血管平滑筋細胞のsmg p21は互いに9個のアミノ酸残基が異なるsmg p21AとーBからなるファミリ-を形成している。(2)smg p21AとーBは共にcAMP依存性プロテインキナ-ゼによって特異的にリン酸化される。(3)血管平滑筋細胞の細胞質にはsmg p21に特異的なGAPが存在する。(4)smg p21はras p21GAPによるras p21のGTPase活性促進作用を抑制する。(5)smg p21の発現は血管平滑筋細胞の分化と共に増加する。さらに、ras p21のC末端のCys残基は翻訳後にファルネシル化され、このファルネシル化がras p21の生理活性の発現に必須であるが、私共は(6)smg p21のC末端のCys残基も翻訳後の修飾を受け、この場合はファルネシル化でなくゲラニルゲラニル化されることを明らかにしている。ファルネシル基やゲラニルゲラニル基はいずれも細胞内コレステロ-ル合成のメバロン酸経路に由来する脂質であり、これらの知見はras p21とsmg p21の活性が細胞内のコレステロ-ル代謝と密接に関連していることを示している。
