局所炎症・免疫反応のレギュレ-タ-としての線維芽細胞

研究概要

線維芽細胞は歯周組織を構成する主要な細胞であり,歯周炎等の慢性炎症病巣の成立・進行・治癒の過程で重要な役割を演じていると考えられる。しかしこれまでの研究は,歯周病関連細菌の菌体成分の線維芽細胞に対する直接的な傷害作用あるいは種々のサイトカインの線維芽細胞に対する作用を調べたものが多く,イフェクタ-細胞としての線維芽細胞の研究は数少ない。我々は線維芽細胞のサイトカイン産生細胞としての側面に着目し,歯周病の病理機序解明を目指して研究を進めてきた。まず10余株の正常ヒト線維芽細胞を小児歯肉より分離した。これらの細胞に種々のグラム陰性菌より調製したリポ多糖(LPS)を作用させたところ、歯周病原菌と擬されているBacteroides(Porphyromonas)gingiuvalis,Bacteroides intermedius(Prevotella intermedia)なイキン(IL)1βならびにIL6を放出し,細胞結合性IL1αを発現することが明らかになった。また腫瘍壊死因子(TNF),インタ-フェロン(IFN)ーβ,γなどで前処理された線維芽細胞はLPS刺激に応じて,より強く細胞結合性IL1αを発現することを証明した。さらにLPS刺激によるIL1αの誘導には内在性のIFNーβが関与していることを示唆する所見を得た。これらの知見は線維芽細胞が複雑なサシトカインネットワ-クの要めの位置に在って,局所炎症・免疫応答の調節に寄与していることを示唆している。また歯周病関連微生物の侵襲を受けた歯周組織では、サイトカインネットワ-クに乱れが生じ,線維芽細胞が過剰にサイトカインを産生し、組織破壊を導びいているとも考えられる。今後、IL8などの他の炎症性のサイトカインと歯肉線維芽細胞との関係なども検討し,歯周組織のサイトカインネットワ-クの破壊と歯周病発症との係わりをさらに明らかにしたいと考えている。
