培養ヒト歯根膜細胞及びラット頭蓋冠由来骨芽細胞の機械的外力に対する影響

研究概要

各種細胞の培養法に関しては、先ずヒト歯根膜細胞について、その培養法を確立し、この細胞が経時的にProstaglandin E(PGE)を産生すること、またその産生能はCytokineであるInterleukin 1(ILー1)やTumornecrosis factorによって亢進することが判明した(研究発表1,2)。しかし歯根膜細胞はprimary caltureである為、個体間格差は無視できない。そのため4人の患者より抜去した第1小臼歯及び第3大臼歯(上下左右を含む)合計6本を用い同様の実験を行ったところ、歯種、性別に関係なく、継代4代目〜7代目でほぼ安定したデ-タを示した(研究発表3)。またラット頭蓋冠由来の骨系細胞は、その培養法もすでに確立され研究代表者によってル-ティ-ンに用いられているが、primary cultureである為歯根膜細胞同様、base levelに差がみられた。そこで今回はcloningされたosteoblastic cellであるMC3T3ーE1細胞を用い、上記の実験を試みたところ歯根膜細胞同様ILー1の添加による有意なPGE量の産生促進が認められた(研究発表4、5)。さらに上記条件下で培養した歯根膜細胞及びMC3T3ーE1が培養期間中にmedium中に放出する物質の骨吸収能を検索するため上記2種の細胞から成るconditioned mediumを用い^<45>Caの放出及び破骨細胞様細胞の出現をみる実験系を行なったところ、両細胞のconditioned mediumは上記の2つの骨吸収活性ともに有意に促進させこの活性はILー1の添加によってさらに増強した(研究発表5)、次に歯根膜細胞の培養中にPGEの合成阻害剤であるIndomethacinを添加すると、産生されるPGEはほぼ完全にblockされるが、そのconditioned mediumを骨吸収系に用いても骨吸収活性は部分的にblockされるのみであった(研究発表6)。以上より歯根膜細胞はPGE以外に骨吸収活性を有する液性因子を少なくとも1つ産生していることが強く示唆された。
