培養ヒト歯根膜細胞及びラット頭蓋冠由骨芽来細胞の機械的外力に対する影響

研究概要	ヒト歯根膜由来細胞(PDL cells)は平成2年度までに確立された方法を用いて継代培養を行いほぼ安定したデ-タが得られる4〜6代目の細胞を、新生Wistar系Rat頭蓋冠由装骨系細胞(bone cells)はすでに確立されている方法を用いて採取し実験に供した。細胞は8×10^5cells/mlの割合で0.3%コラ-ゲンゾルに浮遊させ、14×14mmのナイロンメッシュに支持された厚さ約1mmのコラ-ゲンユニットを作製した。このユニットを今回行開発を行った伸展力反復負荷装置にセットし、PDL cellはDulbecco's modified Eagle Medium(DMEM)(20%ウシ胎児血清:FBS含有)bone cellはEagle's MEM(10%FBS含有)培地にて24時間前培養を行った。機械的伸展(10、15、20%)は1Hzにて行いこの周期的な変形を1時間のうち15分間、間欠的に加え、24ー27時間観察を行った。培養48時間において機械的外力を負荷した両細胞に有意なDNA量の増加が認められた。しかしその伸展変形量の条件は、PDL cellでは15%、bone cellでは10%と異なり、至適条件の違いが示唆された。また骨系細胞では有意なALP活性の低下が認められ、機械的外力は細胞に対し増殖傾向を示すがその分化の程度は低下してゆく傾向が認められた。また歯根膜細胞においてはPGE_2(10^<ー6>M)存在下で、外力負荷時と同等かわずかにそれを上回るDNA上昇作用があり、さらに両者の併用により相加的効果が発現したことにから両者のDNA上昇作用はそのメカニズムに違いがあることが示唆された。組織学的所見において細胞は3次元的に四方に配列されていた。伸展変形を加えた細胞は伸展変形により形態変化を起こしていることが観察された。また、PDL cellは伸展の程度に応じて細胞が伸展方向に伸ばされているのが観察された。本実験モデルを用いることにより生理的状態に近い3次元的な力を細胞に加え形態的な変化が認められた。また機械的外力を研究するモデルとして有用であることが示唆された。
