代謝系の自己組識化に基づく細胞行動における位置情報の生成

研究概要

刺激応答および行動にともなう細胞内化学成分と細胞内構造形成の時間・空間的変動をしらべた。
1.化学パタ-ンの形成と細胞骨格の再編成との対応。
粘菌の生理的条件下における細胞骨格(複屈折繊維)形成のダイナミックスを、マイコン画像処理法を開発して解析した。走電性発現に伴って、陽極側で複屈折繊維が増加し、陰極側へうねりを伴いながら減少していった。このとき、ATP濃度の低いところで細胞骨格は多く形成されることがわかった。したがって、自己形成されたATP濃度分布に従ってアメ-バ運動がコントロ-ルされていると考えられる。
2.細胞行動に伴うリン脂質の時間・空間分布。
細胞膜の重要な構成成分であるリン脂質組成の刺激応答や行動発現に伴う変動を調べた。フォスファチジン酸は、忌避刺激により上昇し、誘引物質では減少した。一方向に移動している粘菌では、前端部で低く後部で高くなりるという極性分布をとることがわかった。
3.Cellーfreeシステムでの新しい化学振動の発見。
粘菌ホモジェネ-トのリン脂質組成が振動していることがわかった。
4.紫外線および各種ホルモン刺激によりヒト表皮角化細胞のCa^<2+>濃度が上昇することがわかった。
以上、細胞は非線形非平衡条件にあるダイナミックな化学反応系として、化学振動や空間パタ-ンと言った時間空間秩序を自己組織化しており、これが生物情報処理および位置情報生成と深く関係していることが明かとなった。
