血管のstretch activationを用いた物理情報の受容と応答機構の研究

研究概要	数多くの興奮細胞より成り立つ心・脈管系は液性調節因子の化学受容と応答の場のみならず、血流や血圧などの血行力学因子に代表される物理受容と応答の場でもある。著者は脳および冠動脈における伸展や歪刺激などの物理受容の細胞内機構について、生体工学的方法と薬理学的方法を組み合わせ、特にカルシウムシグナリングの観点から調べた。本研究に於いては1)物理受容の感応素子であるバイオセンサ-の機能特性、2)バイオセンサ-からのフォスフォリパ-ゼCやL型Caチャネルの活性化過程、3)筋原性収縮反応に対する血管拡張薬の作用、等について研究し以下の結果を得た。1)センサ-部位の機能は糖鎖末端部の切断をするシアリダ-ゼ処理では抑制されず、アルキル化剤であるフェノキシベンザミンや連鎖ブドウ状球菌由来アルファ毒素で抑制された。細胞膜のアルキル化や、アルファ毒素による膜の6量体形成に伴う有孔化によって影響を受け易い因子が物理受容に関係する可能性が示された。2)物理受容によるGTP結合蛋白(Ga)を介するフォスフォリパ-ゼCの活性化によるイノシト-ル1、4、5三燐酸(IP_3)の生成が収縮発生に先立ち亢進する。このIP_3がカフェイン感受性および非感受性Ca貯蔵部位からのCa遊離を促進し細胞内Ca濃度の増加に結びつく。また、伸展などの物理刺激はL型Caチャネルの活性化を促進する。その機序について、IP_3や蛋白キナ-ゼCなどの細胞内機能因子によるCaチャネルの活性化や非特異的カチオンチャネルを介する膜の脱分極の促進効果などの可能性が示された。3)筋原性収縮反応は新たな作用機序を有する血管作用薬の検索に応用可能であることを示した。これらの実験結果を国際シンポジウムにおいて発表すると共に論文として公表した。
