細胞刺激応答反応におけるプロテインキナ-ゼC分子群の機能分担に関する解析

研究概要	我々は、プロテインキナ-ゼC(PKC)分子種のcDNAのクロ-ニングおよびそれらの生化学的特性についての検討から、細胞の多様な刺激応答系にはCa^<2+>依存性のcPKC(α,βI,βII,γ)を介する経路と共に、Ca^<2+>非依存性のnPkCεを介する経路が存在することを示唆した。本年度は、新たにマウス表皮cDNAライブラリ-からnPKCηをクロ-ニングし、分子構造的にnPKCεのグル-プに属すると考えられるPKC分子種が当初の予想をはるかに越えて存在していることを示した。現在、nPKCηの生化学的特性について詳細な検討を加えているが、その結果の一部はJ.Biol.Chem.に掲載した。更に、ラット下垂体由来GH3細胞には、Ca^<2+>依存性のcPKCαとβIIおよびCa^<2+>非依存性のnPKCεの少なくとも3種類のPKC関連分子種が発現していることが明らかとなった。これらのうちβIIとεはホルボ-ルエステル処理によってほぼ完全に細胞から消失するが、αの量的減少は僅かであった。他方、ホルボ-ルエステルと類似の刺激応答反応をこの細胞に引き起こすことが知られている視床下部ホルモンのTRH(Thyrotropinーreleasing hormone)処理によっては、もっぱらεのみが消失した。これらの結果は、Ca^<2+>非依存性のnPKCεが生理的物質であるホルモンの情報伝達系に関与していることを強く示唆するものである。更に、ホルボ-ルエステルとTRHでは、PKC分子種に対する活性化能が異なることを示めしている(BBRCに提載)。現在、3種類のPKC分子種がGH3細胞の種々の情報伝達系にどのように関与しているのかを検討するために、各PKCの人工突然変異体を組み込んだ発現ベクタ-を作成している。また、マウスのnRKCδとηに対する抗体が得られたので、これらの分子種の発現についてイムノブロット法にて検討をする予定である。
