シロイヌナズナ突然変異体を用いた光合成遺伝子発現の制御遺伝子の解析

研究概要

光合成遺伝子の発現制御系を解析するためには,突然変異体を用いた方法論が有効であると考えられる.分子遺伝学的解析に必要ないくつかの条件を満たしているシロイヌナズナ(Arabidopsis thaliana)を実験に供した.
1.光合成突然変異体の生育条件および赤色螢光による一次選抜法の確立
ブドウ糖あるいはショ糖を含む寒天培地(Gamborg B5)を用い、,3ー(3,4ーdichlorophenyl)ー1,1ーdimethylurea(DCMU)により光合成電子伝達系を阻害したA.thalianaが生育することを見いだした.これら光合成活性低下植物体は,長波長紫外線照射により強い赤色螢光を発した.
2.二次選抜のためのレポ-タ-遺伝子のA.Thalianaへの導入
赤色螢光により選抜される光合成突然変異体の中から光合成遺伝子発現の制御遺伝子の突然変異体を効率よく選抜するために,A.thalianaの変異原処理に先立ち,レポ-タ-遺伝子のA.thalianaへの導入を試みた.光合成遺伝子としてリブロ-スー1,5ー二リン酸カルボキシラ-ゼ/オキシゲナ-ゼ(RuBisCO)遺伝子(rbcS)およびクロロフィルa/b結合タンパク質遺伝子(cab)に着目した.A.thaliana ecotype Columbia(Somerville)の葉DNAを鋳型として,クロ-ニングのための制限酵素部位を導入した合成プライマ-および校正活性3′→5′proofreading exonuclease activity)を有するVent DNAポリメラ-ゼ(New England BioLabs)を用い,rbcSー3Bおよびcablのプロモ-タ-とその上流を含むDNA断片(それぞれ約1.5kbp)をDNAポリメラ-ゼ増幅法(PCR)により調製した.これらDNA断片の下流にレポ-タ-遺伝子としてβーglucuronidase(GUS)あるいはchloramphenicol acetyltransferase(CAT)遺伝子を置き,A.thalianaへの導入を試みた.
