スピロヘ-タのリボソ-ムRNA遺伝子の構造解析と系統発生、生物進化に関する研究

研究概要

今年度は、まず非病原性T.phagedenisに対するリボソ-ムRNA遺伝子を含むゲノムDNA断片をラムダ・ファ-ジEMBL3にクロ-ニングした。このDNA断片はリボソ-ムRNA遺伝子を16Sー23Sー5Sの順に含んでいた。さらにT.phagedenisおよび梅毒トレポマネT.pallidum NicholsのゲノムDNAへのリボソ-ムRNA遺伝子プロ-ブを用いたサザン・ハイブリダイゼ-ションから、病原、非病原トレポネマの何れも少なくとも2組のリボソ-ムRNA遺伝子をその染色体上に持っていることを示した。
次に、ライム病ボレリアBiorrelia burgdorferiのDNAを調製後、サザン・ハイブリダイゼ-ションにより遺伝子解析を行い、23S遺伝子と5S遺伝子は非常に近接して存在し、同一ゲノム上に3.4Kbの23S/5Sの2回繰り返し構造を持っていた。16Sリボソ-ムRNA遺伝子は1つが23S/5Sから離れて上流に位置した。一方、同じボレリア属でも回帰熱ボレリアでは繰り返し構造は認められなかった。
前年度、本年度の研究において、トレポネマのリボソ-ムRNA遺伝子は殆どの細菌で見られる16Sー23Sー5Sのいわゆるコンセンサス構成であること、ライム病ボレリアは16Sリボソ-ムRNA遺伝子が23S/5S遺伝子から離れて存在すること、レプトスピラでは3種のリボソ-ムRNA遺伝子がそれぞれ独立して存在することを見出し、これらスピロヘ-タにおけるリボソ-ムRNA遺伝子の発現機序の異なることなどを明らかにした。各種スピロヘ-タは互いに類似した独特の特徴的形態を持っが、生理化学的には属間でかなりの隔たりのあることが知られている。その相違が今回のリボソ-ムRNA遺伝子構成の研究で遺伝学的にも裏づけられ、細菌界における位置と分類の見直しの必要性を示した。
