細胞内寄生原虫感染による自己免疫応答惹起機序の解明

研究概要	トキソプラズマ原虫感染によって抗HSP70自己抗体産生が誘導され、この抗HSP70自己抗体がB-1細胞によって産生されることを既に明らかにした。このトキソプラズマ感染によるB-1細胞の活性化はトキソプラズマ由来ストレスタンパク70(TgHSP70)であることを明らかにした。さらにTgHSP70がB細胞膜上にあるToll-like Receptor(TLR)4を介してB細胞にシグナルを誘導し、さらに非MyD88(おそらくTIRAP)を介したシグナル伝達系により活性化していることを明らかにした。産生された抗HSP70自己抗体の免疫生物学的意義についてモノクローナル抗体を作成し解析した。その結果、抗HSP70自己抗体は宿主免疫防御反応を抑制することが示され、トキソプラズマの宿主免疫反応のエスケープ機序のひとつであることが明らかになった。さらに、トキソプラズマ感受性マウスC57BL/6では抗TgHSP70自己抗体が感染後4-8週にかけて抗体親和性の亢進(immunoglobulin affinity maturation)が起こることを明らかにした。一方、トキソプラズマ抵抗性BALB/cマウスではB-2細胞により産生される抗TgHSP70抗体の親和性亢進が見られ、この抗TgHSP70抗体が宿主の免疫応答の一因として防御反応に寄与することが示された。これらのことは、従来より細胞内偏性寄生原虫であるトキソプラズマ感染に対する生体防御反応は主に細胞性免疫であるといわれているが、B細胞が同時に宿主免疫反応に大きな役割を演じており、さらにトキソプラズマTgHSP70およびTLR4がこの原虫-宿主相互作用の機能分子として役割を演じており、獲得免疫と自然免疫をつなぐ機序になっているものと考えられた。さらには、トキソプラズマ感受性マウスC57BL6マウスにおける抗体自己HSP70自己抗体産生B-1細胞と抵抗性マウスBALB/cマウスにおける抗TgHSP70抗体産生B-2細胞がトキソプラズマ感染後、脾臓やリンパ節において免疫グロブリンの構築発現に必須不可欠であるRAG1,RAG2遺伝子が活性化されることを明らかにし、RAG遺伝子と抗HSP70自己抗体の親和性亢進と何らかの関与があることを示唆した。
