研究概要

淀井らにより報告されたATL由来因子ADFは、ヒトthioredoxin(TRX)のホモロ-グであることが明かとなった。thioredoxinはバクテリアにおいては核酸代謝に関与し、dithiol基を介した還元作用のある補酵素として研究されてきた(A.Holmgren)。また、ADF/TRXはHTLVーI、EBVによるトランスフォメ-ションにより異常発現していることが明らかとなっている(淀井、T.Tursz)。近年T.CullenらによりJun/Fosなどの転写調節因子の活性化、あるいは鉄結合蛋白のmRNAに結合する蛋白の活性化が酸化還元により調節されているという報告(R.Klausner)が見られる。また、還元因子Rafー1の発見がなされた(T.Curran)。蛋白のチオ-ル基の酸化還元反応(redox regulation)が、従来の蛋白質燐酸化を介したシグナル伝達系とは異なった新しいシグナル伝達経路として注目されている。ADF/TRXが、in vitroでNFーκB様蛋白のDNA結合能を増強し(淀井、新井)、しかも細胞内においてILー2R/p55遺伝子の転写を活性化した事より、生体内でADFが転写活性因子の調節タンパクとして作用することが明らかとなった(淀井)。また、HIVの増殖制御機構にも酸化還元機構を介したメカニズムが関与していることが示された(岡本、LA.Herzenberg、淀井、高月)。ADFがTNFや過酸化水素による細胞障害の抑制作用およびラジカルスカベンジャ-活性を示す知見を得ており(淀井)、また、酸化ストレスによりADF/TRX遺伝子が発現誘導される可能性が示されている(淀井)。Bauerleらは細胞外からの酸化ストレスが転写調節因子NFーκBの活性化を行うという報告しているがその機序は明確ではない。また、Storzらは大腸菌において酸化ストレスにより誘導される遺伝子群を報告している。次年度は細胞活性化機構を解析するために細胞内外における酸化還元状態と転写因子活性化の詳細な解析が重要と考えられる。さらに、酸化ストレスとの関連で細胞内シグナル伝達機構を解析する必要があると考えられる。
